    Polymorphisms of two neuroendocrine–correlated genes associated with body weight and reproductive traits in Jinghai yellow chicken

    In this study, insulin-like growth factor binding protein 2 (IGFBP-2) and signal transducers activators of transcription 5b (STAT5b) gene were studied as candidate gene associated with body weight and reproductive traits of the Jinghai Yellow chicken. Single nucleotide polymorphisms (SNPs) of the IGFBP-2 and STAT5b genes were examined in both Jinghai Yellow chicken and three reference chicken populations by PCR-SSCP. Two SNPs (T3746TG and CC3753TT) were detected in the IGFBP-2 gene. One SNP (C8066T) was observed in the STAT5b gene. For primer 1, the general linear model analysis showed that Jinghai yellow chickens with FF genotypes had significant effect on hatch weight, egg weight at 300 days and body weight at 300 days than those of the EF genotype and had significant effect on body weight at 8 weeks of age than those of the EE genotype (P < 0.05). For primer 2, Jinghai yellow chickens with CT genotype had significant effect on hatch weight and age at first egg than CC genotype and TT genotype respectively (P < 0.05). SNPs in both IGFBP-2 and STAT5b genes had significant effect on body weight and reproductive traits of the Jinghai yellow chicken than those with either SNP alone. These SNPs may be served as a potential genetic marker for growth and reproduction traits evaluation of the Jinghai yellow chicken.Key words: Jinghai Yellow chicken, IGFBP-2 gene, STAT5b gene, economic traits, polymorphism

    Bio-Inspired Aggregation Control of Carbon Nanotubes for Ultra-Strong Composites

    High performance nanocomposites require well dispersion and high alignment of the nanometer-sized components, at a high mass or volume fraction as well. However, the road towards such composite structure is severely hindered due to the easy aggregation of these nanometer-sized components. Here we demonstrate a big step to approach the ideal composite structure for carbon nanotube (CNT) where all the CNTs were highly packed, aligned, and unaggregated, with the impregnated polymers acting as interfacial adhesions and mortars to build up the composite structure. The strategy was based on a bio-inspired aggregation control to limit the CNT aggregation to be sub 20--50 nm, a dimension determined by the CNT growth. After being stretched with full structural relaxation in a multi-step way, the CNT/polymer (bismaleimide) composite yielded super-high tensile strengths up to 6.27--6.94 GPa, more than 100% higher than those of carbon fiber/epoxy composites, and toughnesses up to 117--192 MPa. We anticipate that the present study can be generalized for developing multifunctional and smart nanocomposites where all the surfaces of nanometer-sized components can take part in shear transfer of mechanical, thermal, and electrical signals

    Evidence for a Type-II band alignment between cubic and hexagonal phases of GaN

    The study of photoluminescence spectra of a series of thin, undoped, hexagonal GaN films containing cubic GaN inclusions grown by molecular-beam epitaxy on 6H-SiC was presented. It was shown that an emission peak at ∼3.17 eV in thin, hexagonal GaN films exhibits behaviors typical of a spatially indirect transition.     Coralline Sr/Ca and Mg/Ca thermometer for the northern South China Sea: calibration and primary application high resolution on high-resolution SST reconstructing

    利用全谱直读等离子体光谱(ICP2AES) 的分析方法精确分析了南海珊瑚的SrPCa 和MgPCa 比值,结合实测表层海水温度(SST),标定了海南岛南部三亚海域和西沙海域两个滨珊瑚的SrPCa 和MgPCa 温度计。在此基础上,尝试对两个南海北部全新世时期的珊瑚进行SST记录重建。结果显示约540aB.P. (小冰期) 西沙海域夏季月均SST 较现代低约1℃,而约6 500aB.P. (大暖期) 海南岛三亚海域夏季月均SST则高出现代1.0~1.5℃。The method for precisely and simultaneously measuring the coralline Sr/Ca and Mg/Ca ratios was established using inductively coupled plasma atomic emission spectrometry (ICP-AES). Using this method, the high-resolution Sr/Ca and Mg/Ca ratios of two Porites lutea from Sanya, South Hainan Island and Xisha Islands were measured. By comparing to the instrumentally measured sea surface temperatures in these two areas, the coralline Sr/Ca thermometer and the Mg/Ca thermometer were calibrated. These two thermometers can provide SST records with an error bar < 0.2℃, and they are suitable for high-resolution SST reconstructions in these areas. Based on these two thermometers, two short SST records were reconstructed from two Holocene Porites corals of the northern South China Sea. The results indicated that the monthly summer SSTs in Xisha Islands at about 540 years ago (the Little Ice Age) were 1℃ lower than that at present, and the monthly summer SSTs in Sanya, southern Hainan Island at 6 500 years ago (the Megathermal) were about 1℃ to 1 5℃ higher than that at present.published_or_final_versio

    Increased expression of cyclooxygenase-2 in first-degree relatives of gastric cancer patients

    Aim: To study the expression of cyclooxygenase-2 (COX-2) in human gastric cancer tissues and their paired adjacent mucosa, as well as mucosa from gastric antrum and corpus of the first-degree relatives of the recruited cancer patients. Methods: The expression of COX-2 mRNA in 38 patients with gastric cancer and their 29 first-degree relatives and 18 healthy controls was assessed by the real time RT-PCR. The expression of COX-2 protein was determined by Western blot. Results: A marked increase in COX-2 mRNA expression was found in 20 of 37 (54%) cancerous tissues compared to their respective paired normal mucosa (P<0.001). Interestingly, increased COX-2 mRNA expression was also found in mucosa of the corpus (6/29) and antrum (13/29) of their first-degree relatives. Increased COX-2 mRNA expression was more frequently observed in the antrum biopsies from cancer patients than in the antrum biopsies from healthy controls (P<0.05). In addition, 3 of 23 (13%) patients with atrophic mucosa and 6 of 35 (17%) patients with intestinal metaplasia showed increased COX-2 mRNA expression. Furthermore, COX-2 expression increased in H pylori-positive tissues, especially in antrum mucosa. Conclusion: Increased COX-2 expression is involved in gastric carcinogenesis, and may be necessary for maintenance of the malignant phenotype and contribute to Helicobacter pylori-associated malignant transformation. © 2005 The WJG Press and Elsevier Inc.     Hypoxia induces telomerase reverse transcriptase (TERT) gene expression in non-tumor fish tissues in vivo: the marine medaka (Oryzias melastigma) model

    BACKGROUND: Current understanding on the relationships between hypoxia, hypoxia-inducible factor-1 (HIF-1) and telomerase reverse transcriptase (TERT) gene expression are largely based on in vitro studies in human cancer cells. Although several reports demonstrated HIF-1- mediated upregulation of the human TERT gene under hypoxia, conflicting findings have also been reported. Thus far, it remains uncertain whether these findings can be directly extrapolated to non-tumor tissues in other whole animal systems in vivo. While fish often encounter environmental hypoxia, the in vivo regulation of TERT by hypoxia in non-neoplastic tissues of fish remains virtually unknown. RESULTS: The adult marine medaka (Oryzias melastigma) was employed as a model fish in this study. We have cloned and characterized a 3261-bp full-length TERT cDNA, omTERT, which encodes a protein of 1086 amino acids. It contains all of the functional motifs that are conserved in other vertebrate TERTs. Motif E is the most highly conserved showing 90.9–100% overall identity among the fish TERTs and 63.6% overall identity among vertebrates. Analysis of the 5'-flanking sequence of the omTERT gene identified two HRE (hypoxia-responsive element; nt. – 283 and – 892) cores. Overexpression of the HIF-1α induced omTERT promoter activity as demonstrated using transient transfection assays. The omTERT gene is ubiquitously expressed in fish under normoxia, albeit at varying levels, where highest expression was observed in gonads and the lowest in liver. In vivo expression of omTERT was significantly upregulated in testis and liver in response to hypoxia (at 96 h and 48 h, respectively), where concomitant induction of the omHIF-1α and erythropoietin (omEpo) genes was also observed. In situ hybridization analysis showed that hypoxic induction of omTERT mRNA was clearly evident in hepatocytes in the caudal region of liver and in spermatogonia-containing cysts in testis. CONCLUSION: This study demonstrates for the first time, hypoxic regulation of TERT expression in vivo in a whole fish system. Our findings support the notion that hypoxia upregulates omTERT expression via omHIF-1 in non-neoplastic fish liver and testis in vivo.     Effect of ZnCdTe-Alloyed Nanocrystals on Polymer–Fullerene Bulk Heterojunction Solar Cells

    The photovoltaic properties of solar cell based on the blends of poly[2-methoxy-5-(2-ethylhexoxy-1,4-phenylenevinylene) (MEH-PPV), fullerene (C60), and ZnCdTe-alloyed nanocrystals were investigated. Comparing the spectral response of photocurrent of the MEH-PPV:C60(+ZnCdTe) nanocomposite device with that of the devices based on MEH-PPV:C60and pristine MEH-PPV, one can find that the nanocomposite device exhibits an enhanced photocurrent. In comparing the composite devices with different ZnCdTe:[MEH-PPV + C60] weight ratios of 10 wt% (D1–1), 20 wt% (D1–2), 40 wt% (D1–3), and 70 wt% (D1–4), it was found that the device D1–3exhibits the best performance. The Power Quality (PQ) problems in power distribution systems are not new but only recently the effects of these problems have gained public awareness. These non-linear loads are constructed by nonlinear devices in which the current is not proportional to the applied voltage. For the harmonic elimination different methods are used, but in this paper a novel fuzzy logic controller for a three-phase shunt active power filter for the power quality improvement such as reactive power and harmonic current compensation generated due to nonlinear loads. The hybrid fuzzy sliding-mode control (HFSMC) approach is proposed such that it can be applied with advantages to both fuzzy and sliding-mode controller. Simulation results are presented to demonstrate the effectiveness of the control strategy. The results are found to be quite satisfactory to mitigate harmonic distortions, reactive power compensation and power quality improvement.Peer reviewedFinal Accepted Versio
